Kajian Sistematis tentang Peptida Parenteral: Instabilitas, Mekanisme Degradasi, dan Strategi Formulasinya

Abstract: The trend of using peptides, short chains amino acids, as therapeutic agents has been increasing in recent decades. Therapeutic peptides usually use the parenteral route systemic delivery system, requiring an aqueous formulation. However, formulation, are often unstable. most stable form peptide is a lyophilized powder. Unfortunately, for tropical and developing countries such Indonesia, well from pharmaco-economic point view, if stable, formulations solution preferred. This systematic literature review provides various degradation pathways potentially experienced by available Indonesia solution. Then present known strategies to prevent based on research. Article were searched PubMed, ScienceDirect, Wiley Online Library, Google Scholar databases. Search results with keywords: OR drug AND stability stabilization pathway found 20 articles that met inclusion exclusion criteria. Results showed aggregation common pathway, followed deamidation, hydrolysis, β-elimination, disulphide exchange. main improve buffer selection and/or pH adjustment, addition antioxidants/preservatives, combination divalent metal ions, avoiding light exposure, storage cold temperature.
